چکیده

AIM To explore the role of complement system in the T cell activation. METHODS OVA(257-264) CTL epitope and C3b(AN42) encoding genes were synthesized and cloned into expression vector pEGFP-N1. COS-7 cells were transfected with the recombinant plasmid pEGFP-N1. The expression of the fusion protein was determined by FACS. Twenty-four hours after transfection, cultural supernatants of the transfected cells were collected and added to Ana-1 cell culture. Laser confocal microscopy was used to detect the binding and presentation of the fusion protein by Ana-1 cells. RESULTS C3b(AN42)-OVA(257-264) fusion protein was expressed in COS-7 cells. The fusion protein could bind to Ana-1 cells and OVA(257-264) CTL epitope was cross-presented on the surface of Ana-1 cells. CONCLUSION The C3b(AN42)-OVA(257-264) fusion protein can express in COS-7 cells and possesses natural biological activity.

Fusion of Clostridium perfringens type D and B epsilon and beta toxin genes and it’s cloning in E. coli

Designing and producing a proper fusion construction is the most important problem of producing large quantities of a properly folded functional protein. This construction should have all necessary components of a real gene. A good designed fusion gene construction could be cloned into a good and suitable host. Clostridium perfringens is an important pathogen of humans and livestock and produce...

Construction of Recombinant Bacmid DNA Encoding Newcastle Disease Virus (NDV) Fusion Protein Gene

Background and Aims: Newcastle disease virus (NDV) is one of the major pathogen in poultry. Vaccination is intended to control the disease as an effective solution nevertheless this virus is a growing threat to the poultry industry. F gene open reading frame (ORF) from NDV is 1650 bp, encoding a protein of 553 amino acids that can induce protective immunity alone. The F glycoprotein on the surf...

Anthrax toxin-mediated delivery in vivo and in vitro of a cytotoxic T-lymphocyte epitope from ovalbumin.

We reported earlier that a nontoxic form of anthrax toxin was capable of delivering a cytotoxic T-lymphocyte (CTL) epitope in vivo, such that a specific CTL response was primed against the epitope. The epitope, of bacterial origin, was fused to an N-terminal fragment (LFn) from the lethal-factor component of the toxin, and the fusion protein was injected, together with the protective antigen (P...

Construction of the Chimeric HSP70 - E7 Vector and Evaluation of its Protein Production

Background and Aims: Since the produced recombinant proteins by molecular genetics techniques commonly have some limitations in the application, chimeric protein are introduced. Chimeric proteins have found widespread application for the study of protein folding, structure stability, function and immunogenisity. Methods: According to the known Immunomodulatory effect and structure of HSP70 mole...
